Marisa Kirisame is a human, magician, and one of most important sub-characters of the series (according to ZUN). Her personality is sometimes described as disagreeable, but in some other titles, straight-forward. Either way she is a very self confident person that likes to stick her nose into every trouble out of curiosity.
During the events of Story of Eastern Wonderland, she made her first appearance as a minion of Mima, to prevent Reimu from reaching her boss, but was defeated. Since then, she has appeared in every title but one of the Touhou series (and some non-Touhou titles too), usually sided with Reimu, but sometimes against her.
She is also known as a hard worker, who's trying to catch up to Reimu with her secret training and magic development. That makes a good contrast to other humans appearing in the series, most of them have some kind of natural born talent (Reimu) or ability (Sakuya). So she is sometimes referred as the "most human-ish human" in the series.
Fun Facts
The characters for Kirisame literally mean "drizzle" or "misty rain". Marisa doesn't make much sense though... the first character means "magic", the second "logic" or "reason", and the third "sand".
Marisa is one of the most frequent characters to appear in the Touhou series, second only to Reimu.

Despite her self-indulgent personality and lack of concern for her fellow man (or most likely because of these things), Marisa happens to be one of the most popular characters to fans, winning a 2005 Touhou popularity contest.
She is known for "copying" attacks from enemies she has fought, namely Non-Directional Laser from Patchouli and Master Spark from Yuka Kazami. Additionally, the green projectiles that she fires as a playable character seem to have been lifted from Reimu.

A magical girl living in the east. She is a girl who is attached to power, confident enough to like showing off her power, and is always looking for someone to fight with and someone to mock.
However, the truth is she is a hard worker and her power is the result of her secret training.
Because of this, she often gets jealous of Reimu who never trains but still exceeds Marisa with her talent alone, and always tries to get in Reimu's way.
Suprisingly, Marisa is very attached to Reimu at the same time, so much that she would die of boredom without her.
Abiding by the fate of a magician, Marisa isolates herself from other people. Although she possesses sharp wisdom, she normally doesn't use it to her benefit. As a being that humanity has left behind, she doesn't get involved with them. Even so, she can be appeased very easily. Among all, Reimu is her favourite thing (?), so she frequently visits the red shrine maiden to play.
Her weapon is magic. She leads a self-indulgent life full of playing.
This girl is also one of the protagonists. These two (Marisa and Reimu) are the heroines of the Touhou series.
As you know, Marisa has a disagreeable personality and, yes, she really is evil in a sense. Although she's human, she remains distant from other humans and doesn't care much for the well-being of humanity overall. It's questionable as to whether she really is human, but she is.

There are many precious grimoires and other magic items she has collected from all sorts of places all around her house. However, she doesn't use them.
Magic is her weapon. For the time being, her aim is to create the elixir of immortality.
Both of her Spell Card types are certainly not lacking in damage. She is the only speed-type character.
"Magic Napalm" is not very effective against moving targets.
Love Sign's "Illusion Laser" continues to hit enemies with its afterimage, so it is easy to continuously damage the enemy, making it quite useful.
Also, "Master Spark" is a powerful bomb that can deal the highest damage with good stability.

It looks like her habit is getting more serious after each year.
Her house, which she rarely invites anyone into by the way, is located in the forests of Gensokyo (a.k.a. Forest of Magic). It is a bit on the smallish side. The interior is extremely disordered, and would cause an avalanche of collected items if an earthquake hit.
When magic items are gathered in one place they tend to interfere with each other, causing their powers to amplify, weaken, or in some cases even develop completely different powers. Due to this fact, this house may hide items that even Marisa herself never imagined possible.
On another topic, the elixir of immortality she had been developing during the winter ended up being too large for consumption. More improvement required.

An ordinary magician. She usually cloisters herself in her house located at the Forest of Magic, where there are almost no visitors, and researches magic.
She is a bit of a sore loser and meanie, but deep down she has a straightforward personality. She is treated like an annoyance wherever she goes, but in fact she often causes the annoyance to begin with.
One of the reasons she stays in the forest is because no one bothers to go there. She doesn't want to be disturbed while she's doing research, and when she wants to meet others she can always go there herself. She's like a send-only cell-phone. However, she causes a disturbance at other places, wherever she goes.
She has good speed and power, but poor melee technique. While her movement is fast, her attacks are slow. She covers for her lack of physical strength with magic, but the amount of enhancement is actually overkill.

You might imagine witches as being indoor-type people, but Marisa is a rather active outdoor-type person. She prefers a lively atmosphere over silence, but still doesn't like to be disturbed when she's studying. The Forest of Magic is the perfect place for her because almost no one bothers to visit her there. Marisa would also like to point out that, "Concealing my half-finished works is NOT the reason I'm living in there."
A bit past midnight, she felt a disturbance ripple through the forest. She had a feeling that something bad was about to happen, and she was right. Many hours later, it was still as dark as midnight, long past the time for dawn, as though the night was everlasting. She left home to investigate this interesting night... and was very surprised when she met the people responsible for the unnaturally long night.
