Playable Character
In the trial version of the game, Reimu Hakurei is the only playable character. For her available arsenal of towers, see Gensokyo Tower Defense: Towers.
Enemy Characters
Each type of enemy character has a normal form, and an enlarged boss form. The boss versions have more hit points, but also give more power when defeated.
Fairies
Basic normal enemies. There are four visually distinct types, but there don't appear to be any significant differences between them.
Evil Eye
Slow, but damage resistant. Damage for any hit against a normal Evil Eye = normal damage - Evil Eye level. (Thus, for example, a level 0 Homing Amulet would be incapable of damaging a level 9 Evil Eye: 5 damage - 9 EE level < 0.) Boss Evil Eyes are still slow, but lack the special damage resistance, thus making them not much of a threat. When targeting an Evil Eye, the Youkai Buster switches to a 3-way shot.
Crow
Fast (but cannot travel over turrets, bookshelves, or other walls, even though they're flying).
Lily White
Lily simply makes a beeline for the exit, flying over any obstacles in her way. Furthermore, normal Lily cannot be hit by Yin-Yang Gems, though boss Lily can. Generally the biggest threat in the game.
Fluff Ball
Individual fluff balls aren't much different than the fairies in behavior...
Great Fluff Ball
However, some of the fluff balls come in pairs. When a pair is defeated, the fluff balls in the pair split apart, and continue on as separate enemies. The force of the splitting can propel the fluff balls across narrow portions of your walls. An unsplit pair reaching the exit does only 1 damage to your health, but a split pair can do 2 damage. Also note that only the initial split grants you any power items and points; defeating formerly joined fluff balls gives you nothing.
